The Influence of Temperature and pH to Catalyze Enzyme’s Work

Tools and Materials
1.    10 reaction tubes
2.    1 rack of reaction tube
3.    Hydrogen Peroxide(H2O2) solution
4.    NaOH solution
5.    HCl solution
6.    Wooden tweezers
7.    Palm-leaf rib
8.    A beaker glass 200 ml
9.    Cotton
10.    Pure water
11.    2 pipettes
12.    1 set of mortar
13.    1 Bunsen lamp

Steps
The experiment using fresh chicken liver to determine the influence of temperature to the catalyze enzyme:
1.    Make an extract by pounding the fresh chicken liver into the mortar, then add the water as enough. Then, sift it.
2.    Pour the extract into the three reaction tubes by using pipettes. Give 20 drops for each tube or as high as 1 cm.
3.    Prepare three reaction tubes which have already been added by 20 drops of H2O2 for each tube.
4.    Then, pour the first H2O2 solution into the fresh chicken liver extract. Cover it with cotton. Prepare the burned palm-leaf rib which leaves the flame in it, after that put in between the interstice of the bubbles (the result of H2O2 reaction) in the reaction tube.
5.    Heat the second fresh chicken liver approximately 1 minute.  Then mix the H2O2 solution into it. Cover it with cotton. Prepare the burned palm-leaf rib which leaves the flame in it, after that put in between the interstice of the bubbles in the second reaction tube.

6.    The third, mix 20 drops of HCl solution and fresh liver extract. Add H2O2 solution. Close it with cotton. Then, add Prepare the burned palm-leaf rib which leaves the flame in it, after that put in between the interstice of the bubbles (the result of HCl reaction) in the second reaction tube.
To examine the catalyze enzyme in fresh cow liver, use the same method as in the fresh chicken liver.
